Aspergillus oryzae
β-galactosidase, part II: Operational stability, productivity, and process economics Article Snippet: Galactooligosaccharides (GOs) are industrially produced by β-galactosidase-catalysed transglycosylation of lactose, where process productivity and enzyme stability critically affect economic performance. this work analyses the influence of reaction temperature on the productivity, operational stability, and economic feasibility of GOs production using an immobilized β-galactosidase from Aspergillus oryzae.. Batch experiments demonstrated a dual effect of temperature, with higher reaction rates achieved at elevated temperatures accompanied by accelerated biocatalyst inactivation and a slight reduction in maximum GOs yield above 40 °c. to avoid extrapolation from accelerated inactivation studies, a novel batch-based method was applied to directly determine biocatalyst lifetime under process-relevant conditions. at 40 °c, the immobilized biocatalyst achieved a GOs yield of 24%, an initial specific productivity of 0.4 kg GOs kg−1 biocatalyst h−1, and an operational lifetime of approximately 600 h, corresponding to a total turnover number (ttN) of 90 kg/kg.. Balancing GOs yield, reactor productivity, and long-term stability, 40 °c was identified as the most suitable operating temperature for potential application in a packed-bed reactor (PBR). a techno-economic assessment of a continuous process employing the immobilized biocatalyst, including downstream processing, was performed and compared with batch reactor scenarios using the free enzyme. the analysis confirms that GOs production based on the investigated commercial enzyme is economically viable over a broad range of production scales, with the immobilized biocatalyst providing a modest cost advantage and clear benefits for continuous operation. |
